چکیده
Ichthyosis with confetti (IWC) is a rare autosomal dominant disorder caused by frameshift mutations in KRT10 or KRT1 gene. IWC characterized widespread ichthyosiform erythroderma and, more notably, concomitant revertant skin patches, also known as natural gene therapy (Choate et al., 2010; Suzuki 2016; Nomura 2018). Although homologous recombination underlies mosaicism IWC, the molecular mechanism how mutant induces genetic reversion remains unknown. cytoskeletal and microfibrillar protein that gives physical strength to suprabasal keratinocytes, leading formation of epidermal barrier. Consistently, nonsense missense cause epidermolytic ichthyosis (EI) shows strong fragility such erosions bullae due its deficiency dysfunction. However, at tail domain show no/mild fragility, but mosaicism. Therefore, supposed have different pathology from EI. In this study, we investigated principles underlying toxicity IWC-causing peptides. Given peptides are composed repetitive arginine-rich regions low sequence complexity, suspected change properties C-terminal triggers distinct pathogenic functions. We found intrinsically disordered form membraneless organelle through liquid-liquid phase separation nucleolus. Our biological experiments specific interaction between various biomolecules provides unique toxic functions using droplet reaction platform, lead repression aggregation, which not observed EI-causing KRT10.

Frequent somatic reversion of KRT1 mutations in ichthyosis with confetti.
Widespread reversion of genetic disease is rare; however, such events are particularly evident in some skin disorders in which normal clones develop on a background of affected skin. We previously demonstrated that mutations in keratin 10 (KRT10) cause ichthyosis with confetti (IWC), a severe dominant disorder that is characterized by progressive development of hundreds of normal skin spots via...

Ichthyosis with confetti (IWC) is an autosomal dominant congenital ichthyosis also known as ichthyosis variegata or congenital reticular ichthyosiform erythroderma. It manifests at birth with generalized ichthyosiform erythroderma or with a collodion baby picture. The erythrodermic and ichthyotic phenotype persists during life and its severity may modify. However, the hallmark of the disease is...
متن کاملEarly immunopathological diagnosis of ichthyosis with confetti in two sporadic cases with new mutations in keratin 10.
Ichthyosis with confetti (IC) is a severe non-syndromic ichthyosis due to heterozygous mutations in the KRT10 gene. The disease manifests at birth with erythroderma and scaling and is characterised by the gradual development of numerous confetti-like spots of normal skin. Diagnosis of IC is frequently delayed until adolescence or even adulthood. We report 2 young children who were first diagnos...
متن کاملThe double outburst of the unique object V 838
We present spectroscopic and photometric observations of the recent peculiar outburst of V838 Mon, carried out at Rozhen and Toru´n observatories. Our data cover a period of three months beginning just before the second eruption. The evolution of the outburst is divided into four phases. The changes of particular spectral features for each of these phases are shortly discussed. V838 Mon is one ...
متن کاملDiscovery of a Unique Extracellular Polysaccharide in Members of the Pathogenic Bacillus That Can Co-form with Spores.
An exopolysaccharide, produced during the late stage of stationary growth phase, was discovered and purified from the culture medium of Bacillus cereus, Bacillus anthracis, and Bacillus thuringiensis when strains were grown in a defined nutrient medium that induces biofilm. Two-dimensional NMR structural characterization of the polysaccharide, named pzX, revealed that it is composed of an unusu...
